Since you asked here's the nutshell version.

At this point in the show they are beginning to tie characters together from around the world. There seems to be several key items:
1. There is the son of an indian scientist/professor (a professor himself) that is investigating his father's research on people who have this evolutionalry boost - kinda like x-men.
2. There is an agent for some unmentioned agency that is doing the same investigating and has a mind-altering (super-power) assistant. Plot twist here is that his adopted daughter is the cheerleader who is indestructable. He secretly knows her abilities but the show is portraying him as sort of a bad guy.
3. There is an un-introduced bad guy with super powers that is killing off the super-heros and the FBI is onto him.
4. This is where the save the cheerleader thing begins to tie in... There is a guy who can paint the future and already his comic book is being used by another super-hero that came from Japan after realizing he could bend time and space.
The painter has drawn a huge explosion in NYC. The Japanese guy also saw the explosion in a time trip. This guy from japan (within a time paradox in the show) has gone back to tell the most important character - whose power seems to be that of whichever superhero he is with - that in order to save the world he must save the cheerleader who apparently meets her demise in one of the painters paintings.

I suggest you who haven't been watching it since it started get out there and start downloading the shows!!!

OK here is the list of "heroes" we know about:

Isaac: Can paint the future, but only when he is high.

Nathan: politician that can fly, and as evidenced in a past episode has known about his power for a while.

Peter: Nathan's brother who has "dreamed" he can fly. Who also flew (but while around his brother). He also met up with Isaac (Peter is dating Isaac's ex girlfriend) in Isaac's place and painted the rest of a picture. Peter is kind of like Rogue in xmen but doesn't hurt or have to touch them.

Claire: the cheerleader. "indestructible". has filmed herself having accidents and shows her heal rapidly. She has told her friend (well, her friend has videotaped her 'accidents' and her healing herself) about this and no one else. Her adopted father knows about the tape (stole it). When her friend "found it under his bed" she took the tape and put it in her bag. Her brother took it out and watched it. She is swearing him to secrecy (I don't think that will last, and might actually put her in danger when he does talk).

Nikki: Girl who's mirror image does things differently. Her mirror image (i.e. doppleganger) is extremely strong, angry, and uncontrolled while Nikki is unsure, and 'weak'. Last episode we found out her dopplegangers name is Jessica. Mother of Micah. We can tell when it is Jessica because of a tattoo on her right shoulderblade.

DL: Nikki's husband, who has been in jail for stealing, though he claims he is innocent. A few episodes ago we find out that he IS innocent, and that Jessica is the one who stole the money. DL can "phase" (like kitty pride) through objects. Father of Micah.

Hiro: Oriental dude, who is FUNNY. He cannot speak much English. He can stop time, rewind it, etc. He met up with Nathan a few episodes ago and saw Nathan fly to the ground. Future Hiro (with sword and speaks fluent English) meets up with Peter and tells him to "save the cheerleader, save the world".

Sylar: This guy can force people to try to kill themselves, as well as have super strength. He's also ripped the skulls off of people and took their brains. The FBI (who took Matt, see below, into their confidence after finding out his abilities) are trying to find him.

Matt: Police officer who can read minds. How he found a little girl, has worked at mending fences with his wife (who he now suspects has cheated on him, with his coworker buddy).

Horn Rimmed Glasses: Claire's adopted father who knows Claire's ability (though she doesn't know he knows) as well as kidnapped Matt. Tried to kidnap Nathan but Nathan flew away. NOT sure if this guy is a good guy or a bad guy. Rumors say he is actually a good guy.

Haitian man: Horn Rimmed Glasses guy's sidekick. He can erase parts or all of someone's memory.

Micah: Highly intelligent little boy (Nikki and DL's son) that we just found out can "fix" mechanical objects (a phone that was out of order he used to call his mom. when Jessica picked up the phone he knew immediately that it wasn't his mom but her alternate personality).

Radioactive dude: OH YEAH. this guy. I'm betting he is the one that will set off (or BE) the nuclear explosion that will happen in a few weeks. I believe Hiro said they have five weeks to stop the explosion?

Mohinder Suresh: Son of a indian scientist/professor that is investigating his father's research on people who have this evolutionalry boost. His father's place had a HUGE wall of a map with thumbtacks (I'm assuming are pointing to where our "heroes" are.). When he first went there there was someone in the apartment (that we find out is HRG). Also met up with HRG while taking him for a cab ride (Mo is a cab driver). HRG said some strange things that made Mohinder run away from him.

I think I've got them all so far.

I love this show.

The next two weeks will take us up to the event. "Save the cheerleader, save the world" I CAN'T WAIT!

My guess in all of this is... she needs to survive, and Peter will be the one to save us because of his ability. With Claire to help him heal, Nathan to help him fly, Hiro to possibly go back in time if needed, DL to phase, etc, he can tap into their powers and stop the event that causes the explosion.
