Sure. It is easy. There is an access door under the carpet on the hump. Remove one screw, then that access door. Remove two screws on the transmission and remove that access door. Hook the new band onto the old band and pull them through. The down side is that a rag will be required to wipe your hands clean when complete.
On a serious note, the transmission must be removed. My 46RH rebuild is very similar to your 42RE transmission.
At final assembly the transmission must be vertical, as-shown in the image, so that gravity holds the stack orientation until the assembly is complete.
Yank the transmission. Disassemble. Inspect everything. Fix all the problems. Clean, clean, clean. Use a high-quality rebuild kit. Follow the ATSG manual or the FSM. Adjust the clearances to specification during assembly and all will be good.
